We use Chlamydomonas reinhardtii as the chassis organism for biodiesel production. We build two systems in C.reinhardtii. In the first part, we select some key enzymes in metabolic pathway of algae which can improve the biomass and lipid content. In addition, we select HSP70A as the heat shock promoter, which enable algae to express downstream genes most efficiently under outdoor high temperature conditions. In the second part, we inserted a self-flocculating gene behind a blue-light photosensitive promoter. This can be used to control the expression of flocculating proteins in algae when it meets the requirement of harvesting biomass. After physiological function characterization experiments, our system is proved to work properly. Read more.
