In the past few years, laccase has received much attention because laccases are capable of oxidizing phenolic and non-phenolic lignin-related compounds as well as highly persistent environmental pollutants. This makes them very useful for applications involving a variety of biotechnological processes. This includes detoxification of industrial wastewater, such as paper and pulp, textile and petrochemical industries.
Laccase is also valuable as a medical diagnostic tool and as a tool for removing biocides from herbicides, pesticides and certain explosives in the soil. Besides, these enzymes can also be used as catalysts for the manufacture of anticancer drugs, and even as ingredients in cosmetics. Their ability to remove heterogeneous biomass and produce polymer products makes them useful tools for bioremediation purposes. We plan to express the laccase to degradate m-phenylenediamine.
J23100 constitutive promoter
Parts J23100 through J23119 are a family of constitutive promoter parts isolated from a small combinatorial library. J23119 is the "consensus" promoter sequence and the strongest member of the family. All parts except J23119 are present in plasmid J61002. Part J23119 is present in pSB1A2. This places the RFP
downstream of the promoter. Reported activities of the promoters are given as the relative fluorescence of these plasmids in strain TG1 grown in LB media to saturation. See part BBa_J61002 for details on their use.These promoter parts can be used to tune the expression level of constitutively expressed parts. The NheI and AvrII restriction sites present within these promoter parts make them a scaffold for further modification.
BBa_B0015
double terminator
Double terminator consisting of BBa_B0010 and BBa_B0012. This is the most commonly used terminator. It seems to be reliable.
Signal peptide
A signal peptide (sometimes referred to as signal sequence, targeting signal, localization signal, localization sequence, transit peptide, leader sequence or leader peptide) is a short (5-30 amino acids long) peptide present at the N-terminus of the majority of newly synthesized proteins that are destined towards the secretory pathway. If we found the protein(s) which are able to degrade m-phenylenediamine, we would try to secret the protein(s) out of the cell to improve the efficiency of degradation. Here is signal sequences list [1]:

We use these signal peptides infusion with GFP: LPPGFP, LTBGFP, OmpAGFP, OmpCGFP, OmpFGFP, OmpTGFP, PelBGFP, PhoAGFP, PhoEGFP.
B0034-RBS-A, B0034-RBS-B, B0034-RBS-C, B0034-RBS-D
These RBS sequences are a little differnece to BBa_0034. As the sacr between BioBrick RFC_10 and typeIIS assembly are different and may effect the protein expression, we constructed the four RBS to measure the expression strength.
