2.We investigated the actual treatment environment of industrial wastewater and obtained sewage samples in Hangzhou wensli co., LTD.
3.We interviewed the staff of the Hangzhou Eco-environment Bureau and learned about the current status of wastewater treatment.
4.We interviewed Mr. Hu Hong, deputy director of the Experimental Teaching Center of Environment and Resources of Zhejiang University. and got a lot of opinions about industrialization and project transformation.
Gold Medal
a. 「Integrated HP」
1 .We conducted a questionnaire survey to understand the public's understanding of azo wastewater and determined our project.
2.We interviewed the staff of the Hangzhou Eco-environment Bureau and learned about the current status of wastewater treatment. And we understand the current processing methods and try to make up for the shortcomings of existing methods in our projects.
3.We interviewed Mr. Hu Hong, deputy director of the Experimental Teaching Center of Environment and Resources of Zhejiang University. and got a lot of opinions about industrialization and project transformation.
4.Therefore, we have designed an experimental scheme that is more suitable for industrial conditions.
b. 「Model」
We used a mathematic model to analyze the relationship between the absorbance and dye concentration. After the linear relationship between them was established, we started our experiment. We hypothesized that after transforming the laccase gene, laccases were expressed and the bacteria would absorb and degrade the dye in the cells. Though the bacteria have the ability to degrade the dye, the dye is still mainly catalyzed by laccase. Compared with the experiment results with the model analysis, it is confirmed that our hypothesis is reasonable.
c.[Demonstration]
We considered filter element as a whole enzyme in the filtration system.We designed our physical model based on observations from factory field surveys.
We also obtained printing and dyeing wastewater from actual production in the local textile factory and carried out wastewater degradation in the laboratory.
iGEM Special Prizes
a.[Integrated Human Practices]
There are many printing and dyeing enterprises in our hometown, Xiaoshan. Our aim is to help better solve the problems with printing and dyeing wastewater. In order to better understand the disadvantages of modern sewage treatments, we have done various types of human practices, including professors interview, enterprises visit and field trips. According to them, we have clarified our research direction and further improved our project plan. we designed a machine to help degrade the dye in the wastewater with high efficiency. Also we found that the public rarely knew the composition of waste water and the treatment methods of wastewater, we then participated in some educational activities with the public, illustrating the importance of our project as well as the iGEM.

b.[Education and Public Engagement]
Indicated by our public survey, we found that the public rarely knew about wastewater management and synthetic biology. So we organized public events at Zhejiang University and Hangzhou Low Carbon Science & Technology Museum during this summer, holding speeches, lectures and interactive workshops to teenagers and their families. Additionally, we have established our own WeChat Official Account and posted a series of related articles there, making it easier for public to access the relevant information about sewage treatments. We believe that our work have successfully improved the public awareness of the wastewater management as well as the synthetic biology

d.[Measurement]
We transformed the part BBa_K2684000 (CotA) into the E. coli and re-measured the dye degradation experiments after the linear relationship between absorbance and dye concentration was established.E. coli transferred with cotA could effectively degrade the azo dyes. A computer-based model was applied to analyze the mechanisms of the functional genes. Meanwhile, we simulated the real environment and performed the degradation experiments of the functional genes again. All of the relative data and materials were submitted to the PART's Registry page and our WIKI page.
