Team Leader
Changyu Li, Kexin Gao, and Yuanjing Liu are our team leaders. As team leaders, they were responsible for the coordination of the team and supervised experimental details to promote the experimental process.
Changyu Li contacted with biological companies to buy related reagents.
Kexin Gao is mainly responsible for the human practice, for the division of members and the design of the entire project.
The role Yuanjing Liu played in the experiment was to verify the main parts of experiment, finish the construction of vectors and measurement of CIP.
Experiments
Yuanjing Liu, Qiugu Meng, Qiuyun Huang, Zixin Wang, Jiayi Lu, Xingyu Liao, Yubing Zhou, Yingying Cao, and Jing Yao all contributed to plasmids construction. Additionally, Qiuyun Huang, Zixin Wang, Jiayi Lu, Xingyu Liao, Yubing Zhou, Yingying Cao, and Jing Yao were both responsible for improving the previous parts.
Yuanjing Liu and Qiugu Meng were mainly responsible for experiments in vector construction, ciprofloxacin (CIP) detection and CrpP enzyme function verification.
1.They align the correlation sequences and construct the plasmids responsible for CrpP enzyme production, which are piGEM2019-01 and piGEM2019-02, then transform them into different E.coli strains respectively and simultaneously.
2.They utilize High-Performance Liquid Chromatography (HPLC) to measure CIP’s concentration in drugs and establish the standard curve.
3.They extract crude enzyme and verify its funtion.
4.They finish the purification of CrpP and measure its activity by a coupled enzymatic assay involving NADH oxidation.
Qiuyun Huang and Zixin Wang were mainly responsible for qnrS1 verification and improve a previous part. To verify the function of qnrS1, they compared the growth situation of one E.coli with piGEM2019-01 and another one without qnrS1 in different ciprofloxacin concentrations. Additionally, they improved the part BBa_J364000. They added a surface display protein and a linker to the upstream of GFP, and the GFP would be located on the cell membrane.
Jiayi Lu,Yubing Zhou and Xingyu Liao were mainly responsible for PtisAB verification and improve a previous part.
To verify the function of PtisAB, they induce the E.coli with piGEM2019-01 and another one without piGEM2019-01 by different contentration of ciprofloxacin, and detect the intensity of green fluorescent protein, so they can find out the most appropriate concentration of ciprofloxacin for PtisAB.
They also improved BBa_K2617000 by supplementing more data. For this part, A plasmid containing only the bifunctional enzyme gene (xyn10d-fae1a) was constructed and transformed into E.coli BL21. The enzyme was isolated and purified, and the enzyme activity was measured with xylan and ferulic ester as substrates.
Yingying Cao and Jing Yao are mainly responsible for the kill switch.
Their main work is divided into two parts.
The first part is to supplement the data of part BBa_K2556051. They obtained the corresponding plasmid from the ZJUT-China team and transformed this plasmid into two different E.coli strains to map the growth of the host strain curve to complete the supplement of the data.
The second part is to construct a light-sensitive E.coli lysate plasmid and verify the function to achieve growth control of the engineered bacteria.
Human Practice
Kexin Gao, Linhan Huang were mainly responsible for human practices and Changyu Li also contribute to it.They had made the following contributions: designing questionnaires, conducting interview survey and hosting the meetup.
Model
Jiawei Huo and Hairong Wang were mainly responsible for modeling.They made the following contributions to the project:
1.Experimental arrangement
They helped the experimenters scientifically design the experiment to get the required data. They will use the data to get the optimal conditions for the degradation process.
2.Quorum sensing model
Through the establishment and solution of differential equations, we used this model to predict the relationship between the production of CrpP enzyme and the external concentration of AHL.
3.Device layout optimization model
The idea is combined with human practice to help human practice determine the strategy of the placement of the trash cans and the devices.
Artist
Congyu Xu was mainly responsible for art design. As an art designer, she focused on cooperation and contacted Human Practice, Wiki and experiment. Her work included our team logo, team uniform, team banner, the design of the page and illustrations of Wiki as well as the posters and Slides required for meetings such as Southwest Alliance and CCiC.
Wiki Engineer
Minghao Fang and Jiawei Huo were mainly responsible for the establishment of our Wiki page. During the whole project, they warmly cooperated with art designer and designed a set of Wiki style. They were in charge of coding and debugging. Meanwhile, they collected project content from other members and delivered it to the website and ensured complete display of Wiki.
Hardware
Bojun Hou and Minghao Fang were mainly responsible for the establishment of the hardware model. During the project, they designed models ,purchased selected materials and collected test data.Jiawei Huo provided circuit and theoretical support to combine Mathematical modeling and hardware. In a word, they worked to open up our team's hardware.
Advisor
Zhaohui Zhong
He taught us how to design construction and primer for vector, and checked the sequence of primers whether it’s right or not. He also helped us analyze the reasons for the failure of the experiment and redesign the experimental plan.
Jianzhe Yang
He helped us build teams, learn about iGEM, and understand the concepts of synthetic biology. He also provided advice when we choose topics, and also provided ideas for our human practice.
Zijian Wu
He taught us how to choose the direction of mathematical modeling and the method of modeling. He has provided many effective suggestions for our mathematical modeling and hardware design.
PI
Lixia Tang
She is one of our instructors. She gave us some guidance on modeling. She also gave us a hand on the experiment of protein expression and detection of enzyme activity. Also, she helped us to make the data analysis. Moreover, she came up with a series of valuable advice about the design of wiki and ppt.
Yong Zhang
He is one of our instructors. He guided us to design the whole project and helped us to check the construction strategy whether it’s feasible. Especially, he gave us very professional guidance on molecular cloning. He also helped us to comb the pathway and analyzed the result. What’s more, he gave some useful suggestions on wiki and presentation.
Xuelian Zheng
She is one of our instructors. She gave us a lot of suggestions in details during the process of design and experiments. She helped our team leader to coordinate the team works and had communication with each member-friendly to appease our tension. She also raised some ideas about the style of our wiki, poster, banner and so on.
Juan Feng
She is one of our instructors. Meanwhile, she gave us much theoretical guidance on modeling and taught us the way of using various software to found model. She also gave us precious advice on the HPLC design.
