SAFETY

Our lab is the bio-lab of ECUST. We used reagent from Tolo Bio, Axygen, Genscript, Solarbio, NEB, Sangon Biotech, TransGen, NEB, etc. This project was very safe. We only deal with E. coli competent cell and some plasmid DNA. Most of the thing we do was electrophoresis and PCR. Every experiment was performed under the supervising of the Dr. Li and other lab teachers, which made the experiments very successful and secured. Our lab also has very rigid safety reminders. The tips are as follows:

1. Perform only those experiments authorized by the instructor. Never do anything in the laboratory that is not called for in the laboratory procedures or by your instructor. Work areas should be kept clean and tidy at all times. Unauthorized experiments are prohibited.
2. Do not bring unrelated people, medicines, trial products, samples or personal entertainment items to the experiment into the laboratory. Do not play, eat, drink, smoke or make up in the laboratory.
3. After entering the laboratory, the safety door lock must be closed to prevent unrelated personnel from entering. If the door is not closed in time after use, the penalties will be imposed in accordance with the relevant provisions of the platform instrument safety system.
4. Working in the laboratory must be dressed in work clothes, jumpsuits or isolation suits according to laboratory requirements; if necessary, the laboratory goggles, masks or other protective equipment: rings, earrings, watches, bracelets, necklaces or other jewelry should not be worn in the work area. Do not wear shoes with exposed toes in the laboratory; Do not touch pipette with mouth; Do not put the experimental materials in the mouth.

5. Strictly enforce the isolation of areas of contaminated items from areas of cleansing items. Protective clothing used in the laboratory is prohibited from being placed in the same cabinet as the daily clothing; Do not leave the laboratory wearing laboratory protective clothing, such as going to restaurants, offices, staff lounges or toilets; Documents that need to be brought out of the laboratory must be free from contamination in the laboratory; Do not touch the door handle, elevator button or other areas outside the laboratory with gloves.

6. In principle, the instrument platform does not provide drug storage equipment and space. The reagents that brought by the experimenter have to cleaned and taken away after the experiment. If it is necessary for the experiment to save a small amount of reagents on the platform, which needs to be reviewed and approved by the responsible professor, and detailed registration is required. The purchased biomaterials should be received by laboratory specialists and stored in designated locations, making sure documents biological sample information and storage records.

7. All biological waste must be decontaminated by autoclaving or disinfectant before disposal, and they should be stored in a designated safe place before decontamination or final disposal, usually in the laboratory. Do not accumulate waste and laboratory waste, and filled containers should be transported regularly;Do not discard experimental waste. Do not discard sharps, glass, syringes or needles in regular waste bins.
8. To prevent arthropods and rodents from entering the laboratory, please developing effective rodent and pest control measures. Do not place experimental rats in the laboratory for a long time.
9. In the experiment, the broken, spilled or other emergency conditions of laboratory equipment will affect the performance or safety of the instrument and the health or safety of the person. Please protect people first, then ensure the instrument, and finally ensure the experiment. In the case of using electric heating equipment in the experiment, it must be used under the supervision, and immediately shut it down according to the instrument regulations after use, and then cut off the power supply. When laboratory staffs are exposed to infectious substances, they should find emergency aid or contact a professional treatment institution, and report to the laboratory manager in a timely manner, and record the accident and treatment plan.
10. Keep laboratory aisle, public access and fire emergency access. Do not place organic solvents on the floor near the laboratory aisle. All laboratory personnel should be aware of the location of the fire service, the emergency escape route, and the location of the first aid kit.
11. Experiments at night requires to fill an Overnight Response Information Cards. High pressure experiments and other hazardous experiments should be performed in a proprietary laboratory. A transparent shield should be placed between the operator and the experimental equipment in the event of an explosion hazardous experiment.
12. Procurement, storage, and use of the drugs, easy-to-manufacture drugs or narcotics should follow the corresponding regulations; their wastes should be placed in clearly marked containers and handed over to a qualified waste disposal company for destruction. Containers that loaded with drugs must be processed before disposal.
13. Chemical storage should be classified according to the nature. It is best to place the chemicals in a plastic container when transporting them. Disposal waste must be clearly marked with the type, such as organic waste liquid, water waste liquid, solid waste.
14. After the experiment is completed, fill in the instrument usage record and clean up the items and experimental sites in time.
15. After the laboratory is used, the relevant power supply, corridor lighting and doors must be closed. Otherwise, the penalties will be imposed according to the platform instrument safety system.
16. One must receive school fire safety guidance and training, equipped with qualified fire protection facilities, equipment and personnel.
