Establishment the assembly method of type IIS and connected four fragments in one step.
First, we constructed the plasmid pSB1K3-Odd1-lacZ (the plasmid is collaboration with Shanghai_HS_United)
This plasmid is constructed with a lacZ gene that can be used as blue-white screening. We constructed the following plasmid for information storage, and test the blue-white effciency.
We picked the white clony for PCR, and we found all the samples were not the original plasmid (If yes, the PCR product was about 800 bp).
