We validated BBa_I746909 following the measurement protocol by iGEM for this year. It is a composite part made by team iGEM07_Cambridge in 2008. BBa_I746909 includes very popular T7 promotor part in iGEM history, BBa_I719005, that is used 214 times.It also contains BBa_B0034, strong GFP part BBa_I746916, super well-used(4940 times!!) RBS part and double terminators,BBa_B0010 and BBa_B0012 which is extremely popular as well.

pSB1C3 BBa_I746909 is transformed into BL21(DE3) and cultured on two LB agarose gel plates, Cam 34μg/mL.
After these plates were incubated for 8 hours at 37℃,we added 100 μM and 300 μM IPTG to each plates directly, and
