Parts Overview

iGEM Strasbourg project was concentrated on establishing two aptazyme based three-hybrid systems. For this, we linked the aptazyme to MS2 and PP7 stem-loops which represent a big portion of our basic parts. Next to this construct, we tried to produce fusion MS2-LexAmut and PP7-LexA proteins as the components of one of the proposed bacterial three hybrid systems.
Our team added to registry two different types of aptazymes: anti-theophylline switch-off (cleaves itself in absence of ligand) and anti-guanine switch-on (cleaves itself in presence of ligand). Compared to previously described anti-theophylline structures our aptazymes have 100% cleavage activity in vitro. Moreover, guanine aptazyme is the first switch-on aptazyme in the history of the iGEM registry. Our team also added the sequences of inactive aptazymes which are the perfect control for investigating aptazymes activity.
