Part Collection
Our part collection, namely the Kaiser collection, contains 20 universal usable parts. Each of these parts is tested thoroughly as we used them for all of our constructs, which were stringently assessed. By adapting our parts to the type IIS standard, they enable iGEM teams worldwide to rapidly and precisely assemble their recombinant gene constructs. As we use more fusion sites than the iGEM standard, a higher variability can be achieved, so more Level 0 parts can be assembled into an Level 1 part. The collection provides the perfect requirements to start working with Chlamydomonas reinhardtii, as we have made many promoters, terminators, tags and resistant gene cassettes available. Resistance against the antibiotics spectinomycin and hygromycin are widely used within the Chlamy community and allow a simple selection process for positive transformants, these are submitted as coding sequences (Level 0) as well as already functional Level 1 constructs with a promoter and terminator. Through this approach of designing novel recombinant tools, we want to establish Chlamydomonas as a new chassis in the iGEM community. It is a superior model organism when compared to standard bacteria in that, by virtue of being a eukaryote, it has a short doubling time. Additionally, photo autotrophically and is able to modify proteins post translationally. By making secretion signals available, novel iGEM projects can be easily realised through exporting the protein of interest into the medium.
