Project Design

The initial idea of our project was come up during a brainstorming with our PI. Replacing the antigen-antibody interaction with Ligand-receptor interaction in the PRO PREDATOR system (an improved Trim-away method) were proposed then. Moreover, according to the investigation of our HP work, Type two diabetic mellitus (T2D) is related to hyperglucagonemia. Therefore, glucagon (GCG)-glucagon receptor (GCGR) signaling pathway was chosen to be the target of our synthetic modules.

  • GCGR degrading module were inspired by our iGEM 2018 project
  • Glucose sensing module were inspired by our HP work, Xinyuan Qiu helped us design the hybrid promoter.
  • After careful investigation and broad interview by our HP work , we brainstormed and decided to choose primary mouse hepatocytes as the platform for further test of our designed circuit.
  • Human Practice

  • Meiqi Liu and Zuyu Dai were in charge of HP. Meiqi Liu completed the collaboration with hospital and health centers(Nurse Interview and Visit、Clinical interview questionnaire). She also cooperated with the Association of Popular Science Education of Southern Medical University to popularize knowledge about the use of harmless virus as vehicles for gene delivery in disease treatment. Zuyu Dai made the 2019 HP timeline for the team.
  • Yi Qiang mainly participated in meetings and academic exchanges (regular laboratory meeting, academic knowledge report and exchange, the 22nd scientific meeting of Chinese diabetes society and 2019 iGEM Chongqing Meetup).
  • Chaodong Zhang used crawlers to conduct research on the previous year's entries with Huang Zhenyu.
  • Yiman Deng completed two web surveys--diabetes knowledge assessment and viral acceptance for treating disease.
  • Yuhang Ye is responsible for the diabetes education in cooperation with the Red Cross.
  • Qianqian Tao designed and manufactured GI household poster.
  • Model

  • Jie Cai completed the model construction to evaluate the efficiency of upstream promoter.
  • Yiyun Zhang constructed the model to simulate the effect of glucagon and insulin in regulating blood glucose level.
  • Zhenyuan Wang is responsible for summarizing the results of the entire model part.
  • Yanchen Gou is responsible for the overall model construction, data integration and computer implementation.
  • Our PI Xinyuan Qiu and Instructor Chuanyang Liu helped us with modeling ideas and methods.
  • Wet Lab

  • Jiaxin Ma, Jie Cai, Hanxiao Feng, Yating Long, Xinyu Su and Zuyu Dai completed most of the experiments.
  • Jiaxin Ma are mainly responsible for supervising the progress of the wet lab.
  • Jie Cai、Hanxiao Feng、Yating Long、Xinyu Su、Zuyu Dai are mainly responsible for the characterization, description all parts created by our team this year.
  • Wiki build

  • Zhenyu Huang, Qinrui Jiang and Shengpeng Liang worked on the visual appearance of it.
  • Zhenyu Huang was responsible for building the template of the pages.
  • Qinrui Jiang and Shengpeng Liang added elements to pages.
  • Visual design

  • The Giant Jamboree Banner is designed by Yiman Deng.
  • The adenovirus science guide manual is designed by Chaodong Zhang.
  • Qinrui Jiang and Shengpeng Liang were in charge of upload information to our wiki.
  • The wiki home page cartoon is designed by Qiang Yi.
  • Support

    Wiki support

    • Our advisor Haoxing Yang and Jianfang Wang provided us solutions when we face difficulties in the process of wiki production.

    Human Practices support

    • The Association of Popular Science Education of Southern Medical University has provided a valuable platform and experience for the popularization of gene therapy and virus therapy. Simultaneously, we sincerely thank the Red Cross of National University of Defense Technology and Changsha volunteer organization for providing a favorable publicity platform for our diabetes knowledge popularization, which has truly made us communicated with the public. Last but not the least, We would like to thank the medical staff of the Second Affiliated Hospital of Chongqing Medical University for answering our questions without reservation and providing a strong force for the completion of the project.

    Project support and advice

    • This project was come up with due to a brainstorming and then we discussed the details about it in a group meeting. We designed the experiments under the help of our primary PI Prof. Lingyun Zhu. Besides, Dr. Jingyu Kuang and Dr. Xiaomin Wu helped us to be familiar with the experimental operations. Our primary PI Prof. Lingyun Zhu also provided us a dedicated laboratory and equipments. Our instructor Mr. Xinyuan Qiu offered us the guidelines about conventional experimental techniques and great advises to ensure the smooth development of our project, and helped us to contact other colleges and institutes for collaboration.

    Fundraising help and advice

    • We got experimental funding support from our PIs Prof. Lingyun Zhu and travel funding support from College of Liberal arts and Sciences of NUDT. Our advisor Lu Min was in charge of expenses management.

    Lab support

    • Department of Biology and Chemistry of College of Liberal arts and Sciences provided a biology experimental platform for us.

    Project adviser support

    • Our advisor Jingyu Kuang, Lvyun Zhu and Xiaomin Wu provided us solutions when we face difficulties in the process of proceeding the project.

    • Sisi Xie and Wenying Li guided us how to package adenovirus. Xinyuan Qiu guided us to construct adenovirus vectors.
