OUC-China
We are interested in the case of OUC-China about the riboswitch which is a structural genetic regulatory element that directly regulates the expression of downstream genes through the reaction of small molecules. It has a wide range of ligands, so it has a great potential in the fields of synthetic biology. But the secondary structure of riboswitches is often unstable, so riboswitches cannot be considered a modular component. In order to solve the problem of modularization of riboswitches, their team proposed a theory of modular riboswitches. They use a “Stabilizer” to stabilize the structure of the riboswitch and use a “tuner” to insulate Stabilizer and GOI, which allows for predictable tuning.
In order to verify the feasibility of the modular riboswitch constructed by their team in practice, we need to express our sequence in different experimental environments to explore the stability of this structure. We help them analyze the data measured after 8 hours of incubation. So we can conclude that after eight hours the maximum fluorescence values is added 0.15 mM 2-AP.

Changchun meetup
On July 19th to 20th, we gladly accepted the invitation from Jilin_China and took part in the JLU meetup hosted by Jilin_China and JiangnanU_China in Changchun. Jilin_China, JiangnanU_China, NEU_China, NJTech_China and NEFU_China participated in the meeting. We gave a presentation about our project. We got lots of information such as Nissle bacteria could be used to relieve inflammatory bowel disease, and synthetic biology could be used to treat breast cancer. During discussion, we shared our opinion of other teams projects and also got practical advice from them.
Brainstorming About Light Control Superiority
In the early stage of the project, we held a video meeting with iGEMer from DUT_China_B to exchange our project ideas and designs online. We think their project is novelty, and the optical required equipment they put forward is a kind of wireless control, penetrability, good control method. They should pay attention to demonstrating the superiority of optical relative to other control means, however the light pollution and other issues which should be considered at the same time, and they also should ensure the only light source during the process of experimental operation.
