GenesmegL : is from the genome of pseudomonas deceptionis, which is found in the arctic soil, it catalyzes methionine to form methanethiol, an important precursor in DMS production. mddA: A newly discovered gene that encodes MddA enzyme, which methylates MeSH to DMS using Ado-Met as the methyl donor. It is found in a variety of DMS producing soil bacteria, which utilizes an alternative pathway that does not produce DMSP, unlike many other DMS producing algae. Our sequence comes from a paper called “A novel pathway producing dimethylsulphide in bacteria is widespread in soil environments” and it comes from pseudomonas deceptionis. Our gene encodes the enzyme phosphoenolpyruvate carboxylase, which catalyzes the addition of bicarbonate (HCO3−) to phosphoenolpyruvate (PEP) to form the four-carbon compound oxaloacetate and inorganic phosphate. It is observed mainly in plants and fungi, and in our case, arabinose thaliana. We use this constitutive promoter, J23100 promoter, from the Anderson promoter family because it has been shown to work reliably in NCHU 2018 team. Moreover, it has been characterized by Yale to be effective in driving gene expression, by adding citrine and measuring the fluoroscence intensity. |
