Parts

Our parts collection consists of thirty two basic parts and fifty five composite parts. All parts were generated during our project. Some of them are related to the PURE cell-free system, while others are application of Toehold switches.

Best Basic Part

EF-Tu protein: BBa_K2916009

We have chosen to nominate EF-Tu as our best basic part. It is an indispensable component in the PURE cell-free translation-transcription system. The main function of the EF-Tu (elongation factor-thermo unstable) is to catalyze the binding of an aminoacyl-tRNA (aa-tRNA) to the ribosomes. It plays a crucial role in the OnePot PURE cell-free system as it accounts for 47% of the total protein concentration in the protein solution. The sequence, apart from the gene encoding, also includes a 6x HIS affinity tag, to allow the purification of the expressed protein.

Best Composite Part

Toehold switch for Bois Noir 2.1 with superfolding GFP : BBa_K2916073

The ultimate goal of our project is to detect specific sequences of the two diseases in question, so for Best Composite Part we nominate Toehold switch for Bois Noir 2.1 with superfolding GFP. This part, although it is quite particular to our project may act as a proof of concept for future iGEM teams that also want to implement toehold switches in their project.
