What are antimicrobial peptide?
AMPs are currently the potential defense against multidrug-resistant (MDR) pathogens that are naturally occurring molecules as the alternatives to the antibiotics. This kinds of molecules have the broad activities to directly kill many organisms for instance bacteria, yeast, and fungi, etc. The peptide is composed of different lengths and different sequences of amino acids. Unlike antibiotics, AMPs have diverse mechanisms against bacteria which make it hard to develop a complete resistance. Generally, the bacterial membrane is composed of a lipid bilayer structure.
JJ01
JJ01 is an antimicrobial peptide which is originally modify from MAP0403, and it has been granted an United State patient (Patient No.: US 9,802,982 B2). As the previous study indicated that although MAP0403 has antimicrobial potency, it has side hemolytic effect that is not suitable for application. According to researches, the higher amphipathic alpha-helical character may afford it to interact with the bacterial membrane, which acts like a detergent and its analogies. In order to solve the side effect, JJ01 was designed to change the amphipathic structure arrangement of MAP0403 to interfere with the bacterial membrane as the first and foremost strategy. After confirmed its antimicrobial activity, JJ01 is found to be able to inhibit Escherichia coli , S. aureus , as well as P. aeruginosa with 5 μM, 5 μM and 10 μM, respectively. The minimal inhibition concentration (MIC 90 ) of P. aeruginosa increased twice, but Escherichia coli and S. aureus still maintain at 5 μM. Fortunately, JJ01 has been proved to significantly decrease the hemolytic activity by break down the amphipathic structure.In conclusion, JJ01 is an effective antimicrobial peptide that has the potential to replace antibiotics.
