With the success of our team Dr. Thermometer for RNA thermometer experiments, we use GFP fluorescent protein as a chromogenic substance, RNA hairpin structure as a reaction principle, and 20 different kinds of RNA thermometer designed by ourselves according to certain temperature (20 degrees Celsius to 40 degrees Celsius). On the other hand, there are still many aspects for improvement based on the experimental period and the summary analysis after the experiments. At present, the amount of fluorescent protein GFP used in the experiment is not much, and the color(green) of product is not so obvious that the operators can notice immediately when GFP is expressed. In order to solve this problem, we are trying to figure out whether other types of fluorescent proteins can be used in this experiment, such as more vivid or easier to express, to deepen the color of the product. Of course, it is easier for users to observe the color change. Secondly, in terms of temperature, our product RNA thermometer can only be used in the range of 20 degrees Celsius to 40 degrees Celsius, so the main task in the later stage is to expand the range of RNA thermometer use, which means that RNA thermometer can be applied to more drugs with higher temperature requirements. Moreover, we are using a stepped temperature setting, a phase every 2 degrees Celsius, and the next step is to be able to control and set any temperature. Considered the humanization, the product form we design is cumbersome, the operation is a little bit troublesome, and the cost is slightly high. If it is not properly operated, it will be wasted, and the effect of the product will not be achieved. If RNA thermometer is put into practice in the future, we will come up with a more convenient, lower cost product form to benefit more people.
