The RocKit



In this project we respect all of the safety guidelines required by the iGEM foundation. Furthermore, we participated in the RIVM Safe-by-Design grant, which brought safety issues in the design of our project to our attention. We even created an infographic in which we demonstrated different safety aspects of our project. Our project will be using existing technology in a novel way to improve our systems detection mechanism.

Manual:

Step 1.
Make YPD plates

Step 2.
Streak-plate the cells on selective YPD plates

Step 3.
Take several colonies for inoculation in YPD media

Step 4.
Induce mutation mechanisms by centrifuging cells down and adding a 2% galactose rich media with target molecule of interest

Step 5.
To detect functionalized receptors use a flow cytometry set-up for single cell quantisation

Step 6.
Replate bacteria that were positively selected on a new YPD selective growth plate.

Step 7.
Inoculate several colonies for Overnight culture

Step 8.
Use Miniprep Kit to extract DNA for sequencing

Step 9.
Upload sequence to the ROCCloud
