A brand new DNA nano-structure is designed so that it is specific to be bound by
the Lactobacillus DNA fragment and the actobacillus itself. As the below graph shown,
Four single-stranded DNAs were used to form the nano-structure, in which it is shown to be a bi-triangular
plane with four “extension strands”. Corresponding base pairs are filled in two
extended strands so that the base pairs are complementary to parts of the target DNAs,
forcing two bi-triangular plane to “stick together“. Another two extended strands, when
bound with each other, could form a stable G-quadruplex in the presence of hemin because
they are guanine-riched sequence.. As stable form of G-quadruplex could enhance the
peroxidase activity, checking the peroxidase activity of the G-quadruplex would then
reflect the number of the target DNA bound with the DNA nano-device, and in turn reflecting
the concentration of target DNA.
The four oligos that are responsible for forming the nano-structure are listed below for reference.
