Parts
Parts Overview
Our project aim is to build a unique underwater adhesive protein toolbox using parts obtained and upgraded from various aquatic organisms known for their stickiness – mussels (mfp3, mfp5, mfp1) and barnacles (rBalcp19k) as prominent examples, and E.coli curli-forming protein CsgA known for its potential cohesive strength. In total, we submitted 24 parts, with 15 basic parts and 9 composite ones. We designed all these parts based on the iGEM Parts Registry and previous research. Some parts are directly obtained by synthesis, and other parts were designed and assembled in our laboratories.

Parts Collection
We modified our parts and combined genes of different organisms so that they provide different levels of adhesion, cohesion and other various characteristics. We also established a remarkable part collection based on the underwater adhesive toolbox using E.coli as chassis. The part collection contains recombinant proteins – CsgA-mfp5, Fp151, rBalcp19k-mfp5, etc. In addition, a recently reported tyrosinase– mTyr-CNK that is used for DOPA modification of recombinant adhesive proteins containing Mfp was involved in the part collection to make these proteins stickier. Together, the components in each part are closely related to each other and all of the parts fulfilled multiple functions and can be further adapted for use in technical/biological fields. Therefore, we have decided to nominate our parts for the Best Part Collection award.
