Our Worldshaper-wuhan team actively engaged in in-depth cooperation with other teams in the 2019 IGEM competition. We have established a very friendly relationship with XHD-WS-Wuhan-A, XHD-WS-Wuhan-B, HZAU-China, and have gained lots of experience and benefits.
Cooperation in experimental technology
1. We helped Wuhan XHD-B team to conduct SDS-PAGE experiments.
Under the guidance of Wang Cheng from Wuhan University, Huang Yiming in our team, taught Guan Zhiyue from XHD-B team to conduct SDS-PAGE experiments, including preparation of SDS-PAGE electrophoresis gel, preparation of protein samples, protein electrophoresis loading and testing, mas blue bright staining, and analysis and processing of results;
2. We help XHD-WS-Wuhan-B team to carry out WB experiment. We helped XHD-WS-Wuhan-B perform WB experiments, including protein electrophoresis, transfection, blocking, incubation of primary antibodies, secondary antibody incubation, and final exposure.
3. troubleshooting:
Since the XHD-WS-Wuhan-B team's SDS-PAGE electrophoresis effect has not been ideal, and the protein expression has been very small, we helped it optimize the experimental protocol, and proposed a protein purification kit to solve this problem.
XHD-WS-Wuhan-B and XHD-WS-Wuhan-A team helped us successfully construct the plasmid for the characterization of the components.When we built our characterization components, we had a lot of problems, which led to the unsuccessful construction of our characterization components. Later, we handed the experimental materials to the XHD-WS-Wuhan-B team and the XHD-WS-Wuhan-A teammates. Finally, the plasmids that characterized the components were successfully constructed.
4. We worked with XHD-A teammates to help XHD-WS-Wuhan-B teammates experiment with their characterization.
iGEM X High School meet up in Hangzhou
iGEM X High School Conference in Hangzhou
On August 2, our team, Worldshaper-Wuhan was invited to attend the iGEM X High School conference in Hangzhou. Two group members including the team leader attended this conference.
This conference was divided into two parts, which were the same as the procedures in Giant Jamboree: presentation and poster display. The whole conference was aimed to help candidates better understand the process and receive constructive suggestions from judges and professors.
The first part is about presentation. After the presentation, judges and professors gave us some advice:
1. We should deliberately choose what to present in the background introduction in order to control our presentation time.
2. We should adjust and improve the introducing sequence trying to deliver a vivid story which could grab audiences' interests.
3. The detection and absorption was better to be designed into one gene circuit, because it was more economical.
4. The portable device mentioned in the end of presentation was highly recommended to made into finished products.
5. More human practice should be conducted like interviewing stuff in the sewage treatment plant and more professionals.
On August 3, the poster-display part was held. Judges came around and asked for the introduction and description of our project. Since we prepared the poster carefully, the whole process went well. Also, some judges gave their valuable advice to us.
1. The flow of poster presenting should be organized in a fluent order.
6. The illustrating images on the poster should contain any other languages. We should be more careful when chose the images (only English allowed).
7. There should be less introducing information, but more design, experiment and human practice parts on the poster.
8. All the team members should fully understand the project.
