Parts Overview

Biobricks are standardized parts, consisting of nucleotide sequences, that can be placed together to build larger, genetic circuits. Each biobrick consists of a prefix and suffix sequence that each recognizes three different restriction enzymes for a total of six restriction enzymes. These enzymes can then be utilized for cloning purposes as the user sees fit. Our project’s main focus was to confer thermostability in the LaSota strain of the Newcastle Disease vaccine through the use of intrinsically disordered proteins (IDPs). Thus, all of our parts are focused on the protein-coding sequence of various IDPs.

Bronze Medal Requirement

For our Bronze medal requirement, we characterized CAHS 94205 (BBa_K2306006), which is referred to in our lab as CAHS D. The sequence we used in our lab (BBa_K3008003) encodes for the same protein (UniProtKB/Swiss-Prot: P0CU45.1), however, the sequence we used is the native sequence from the genome of Hypsibius dujardini as opposed to the E. coli optimized sequence of BBa_K2306006. This is stated on the registry pages for these respective parts. For our characterization, we documented the production and expression of our protein and performed circular dichroism to view the structural composition under different temperatures.

Silver Medal Requirement

For our Silver medal requirement, we characterized the ability of CAHS 107838 to be subjected to a temperature of 121°C without degrading. This is a quality we are looking for as we will be subjecting these to high heat and we need them to protect the NDV vaccine without degrading.
