Synthetic biology seeks to achieve rational design of circuits to give new functions to living cells. To date, most of the work has focused on gene regulation because transcription factors and other nucleic acid interacting proteins can be configured relatively easily to regulate each other's expression. However, many of the natural cellular functions are achieved through protein-level circuits, and circuits that synthesize protein levels can achieve powerful new cell behavioral engineering. Synthetic protein circuits can provide advantages over gene regulatory circuits, including faster manipulation, direct coupling to endogenous pathways, single transcript delivery, and the ability to integrate without genomics. We have collected a number of components that can be used for protein circuits, including those we built this year, and those that have been built before, and hope that these components can help other teams to achieve good results in the synthetic biology of protein circuits.
