Safety


  • During the project, our team worked mainly in the undergrad lab at the Indian Institute of Science Education and Research, Tirupati. This lab is BSL-1 and has two biosafety cabinets.
  • We had a different lab for working with mammalian cell culture which was equipped with BSL-2 safety cabinets.
  • While performing lab work, correct personal protective equipment (PPE), such as nitrile gloves, long pants, and lab coats were worn by all lab members. Safety goggles and mask were used while handling the acid/base.
  • Waste disposal: The biological waste generated was collected separately in biohazard bags. This waste was then treated with 2% bleach and then sterilized by autoclaving before disposal. Cell lines were disposed of by trypsinizing and then treating with bleach. The waste was then collected in biohazard bags for safe disposal. Ethidium bromide waste was collected separately and disposed of appropriately. Organic liquid wastes were disposed separately from aqueous wastes.
  • All team members had undergone a lab safety training program at our institute before the start of the project, for following the safety guidelines of the Institutional Safety Committee of IISER Tirupati.


Organisms:


  • We worked mainly with E. coli strains DH5α, BL21 and JW4276 and Saccharomyces cerevisiae strains CEN.PK and BY4741, which are non-parasitic and non-pathogenic in nature. These organisms belong to Risk Group 1 and require BSL-1 safety procedures. Experiments involving these organisms were carried out in laminar air flow cabinets.
  • Experiments on the mammalian cell lines HT29 and CRL1790 were carried out in BSL-2 safety cabinets by team members trained in mammalian cell culture in a separate mammalian cell culture facility.
  • To prevent unintentional release of GMOs into the environment, all containers containing bacterial cultures were washed with bleach after use and then autoclaved.
