Improve
We attempted to improve the previously existing Part: BBa_K554005 (Recombinant IL-12) developed by UNICAMP EMSE Brazil. The same team used this part previously used in conjunction with HlyA to make it secretory. We took the part and codon-optimized it using the tool from Twist Bioscience for expression in E. coli BL21 and fused it with a similarly optimized YebF tag (Part: BBa_K1610300) for secretion. We expected that this fusion construct would code for a functional YebF-Recombinant IL-12 protein (with FLAG Tag for detection) which would be secreted out of the cell.
We believe that this is a better method as HlyA requires a dedicated secretion system which must be made available, but YebF requires no such systems.
