Here is a list of accomplishments we have made during the course of our project:

Bronze

Requirement What we have done
1 Registration and Jamboree Attendance 10 members of our team have registered for the Giant Jamboree and will be travelling to Boston to attend the event
2 Competition Deliverables
  • Created a wiki page
  • Designed and printed a poster to introduce our project
  • Will be presenting on stage in Jamboree (Nov2 12:00, Room 311)
3 Attributions Attributed all interviewees, collaboration teams, sponsors, as well as our team advisors and supervisors
4 Project Inspiration and Description We are inspired to improve current repressor protein-based biological switches by introducing a CRISPRi and antisense RNA switch system
5 Characterization of an Old Part We characterised old part BBa_R0010 (a LacI promoter) because it is used in our circuitry

Silver

Requirement What we have done
1 Validated Parts / Validated Contribution New parts BBa_K3017061 (CRIPSPRi sgRNA) and BBa_K3017060 (CRIPSPRi asRNA) are characterized together as a functional pair
2 Collaborations We collaborated with a local high school team (HK_GTC) and mentored another local high school team (Hong_Kong_UCCKE)
3 Human Practices We have sought consultation from some synthetic biologists and bioengineers to see if the CRISPRi switch is appealing to them in terms of future development and application. We have also engaged high school students and the public in events like museum exhibitions and summer camps

Requirement What we have done
1 Integrated Human Practices Dr. Sun Fei and Professor King CHOW suggested multiple promoters in our circuit would compete and interfere expression. We conducted an experiment to predict if the situation would happen in our model circuit
2 Model Your Project We built a descriptive model which characterizes the expression of the output proteins in our circuit to evaluate:
  1. the switch's effectiveness, and
  2. the elasticity of variables
