4. Project Inspiration and Description:
We have clearly documented what we found from the research we did about nickel ion pollution problem. Refer to the background research outside of laboratory that inspired our project, we have also documented how we determined our project goal and described how we will address that by biological method on Project Inspiration and Description.
5. Characterization / Contribution:
We added new, high quality experimental characterization data to two existing Bio Brick Parts from the Registry—— BBa_E0032 and BBa_E0034 on Part’s main page.

1. Validated Part / Validated Contribution:
We designed new Composite Parts, and their expression products in yeast can increase the ability to absorb nickel ions from water, so our results proved that these parts are biologically functional parts. More information can be found on parts' main page: BBa_K3126018, BBa_K3126022, BBa_K3126023.
2. Collaboration:
We have participated in two meaningful meetups this year and learned a lot from other teams. We were also glad to have HZAU-China, HUST-China, OUC-China doing the model work for us. More details can be found on Collaboration.
3. Human Practices:
we stepped out the lab and propagated our project to share our idea of addressing the nickel ion pollution by a biological way. We designed a complete chain of Human practices and carefully finish it, we conducted some HP sets (which concludes input of the real world and output by our team). 1. Integrated Human Practices:
We met with experts from academia, industry and government departments, and these activities provided us with improvements in many aspects, which is an unparalleled comprehensive human practice! Details can be found on our Integrated Human Practices page.
2. Improve a Previous Part / Project:
Our project aims to remove nickel ions from water. We found that BBa_K2652006 can express NikABCDE protein which belongs to the ATP-binding cassette (ABC) protein family, and contains five proteins that can transport the hydrolyzed ATP to the transmembrane transport of nickel ions. This year we made an improvement by adding His-tag sequences to both ends of it to optimize it. We clearly documented it and added high quality experimental characterization data to both parts on Part’s main page of BBa_K3126026.
3. Model Your Project:
HUST-China helped us to obtain a basic model of the gene expression. We thoroughly predicted the movement process of nickel ions in water during reaction in a three-dimensional homogenous space occupied by Ni>2+ by making a convection dispersion model. With the help of HZAU-China we made a mathematical model to demonstrate that TgMTP1t2 protein can alleviate the cytotoxicity of nickel ions. OUC-China helped us made a molecular dynamic simulation of the process that nickel ion entering the cell through the NixA transporter protein. In order to predict the efficiency of nickel absorption, we successfully build an equilibrium enrichment model. More details can be found on model page.
4. Demonstration of Your Work:
In order to prove that our whole project can work, we designed and manufactured a “Mini Factory” device. We put the embedded pellets made of engineering yeast into it, simulated our expected work situation, and received relevant data through the app we designed. More details can be found on Demonstrate .
