Prototype Detector
Since the cadmium dependent parts we required were not available in the 2019 iGEM kit, we decided to make a part to test our hardware bioassay and to prototype our future cadmium detector, since it works in a similar process. It has a Constitutive promoter, followed by MerR, PmerT, GFP, PcadA, RFP and lastly, a double terminator.
The MerR acts as a repressor on the PmerT and PcadA. When mercury is present in the environment, the promoters are unrepressed and both GFP and RFP are expressed.
Current Progress
To create our part, we followed a ligation plan.
We began with digesting the MerR, PcadA and RFP parts to prepare for the ligation. This is the result of our digest.
Unfortunately, we were unable to finish the part before the Giant Jamboree date, so we will continue this project next year.
Future Part
The true goal of our project is to eventually create a part that can detect cadmium. Our goal is to have a detector that can express RFP in under 10 hours, to be applicable to the fishermen. This part would be composed of a constitutive promoter, MerR that is modified to be specific to cadmium, PcadA and RFP.
