Project Inspiration Description

Part Collection

The team BOKU-Vienna created a library of BioBricks that are crucial for the development of a new, fast and easy-to use diagnostic tool for Buruli ulcer. The biobricks are compatible with most assembly standards, including RFC10 and RFC1000.
We successfully screened for a riboswitch that will increase transcription of a downstream located gene in the presence of mycolactone, which is the disease-causing toxin of Buruli ulcer. Further, our part-collection includes all biobricks that were used to proof the functionality of our system with a transcriptional theophylline riboswitch.
The designed and constructed system can easily be adapted for detection of various disease-causing agents by just changing the riboswitch-sequence according to the target to generate a visible readout.
This highlights that our genetic device could act as a great starting point for the development of a diagnostic platform for the detection of rare, overlooked and neglected disease-causing agents or even to screen for toxins in the environment.
