Zixuan Zhang and our advisor Rongrong Du offered lots of suggestions and help. In our brainstorming sessions, everyone in the team contributed to improve the project.</p>

She herself finished the construction and verification of reverse transcriptase parts. Yuchang Li was responsible for the construction and verification of the Cre/LoxP system. Fluorescent measurement was conducted by Xidan Miao. Yi Qiao was our major contributor in molecular cloning. Yichen Liu in company with Yi Qiao constructed our improved parts and measured them. Jingyu Cao also helped in molecular cloning. All the people mentioned above together finished molecular cloning. Zixuan Zhang helped us verify our experimental design.</p>

                    <p style="margin: 0">Contact-dependent signaling is critical for multicellular biological
+
                        events, yet customizing contact-dependent signal transduction between
+
                        cells remains challenging. Here we have developed the ENABLE toolbox, a
+
                        complete set of transmembrane binary logic gates. Each gate consists of
+
                        3 layers: Receptor, Amplifier, and Combiner. We first optimized synthetic
+
                        Notch receptors to enable cells to respond to different signals across the
+
                        membrane reliably. These signals, individually amplified intracellularly by
+
                        transcription, are further combined for computing. Our engineered zinc finger-based
+
                        transcription factors perform binary computation and output designed products.
+
                        In summary, we have combined spatially different signals in mammalian cells,
+
                        and revealed new potentials for biological oscillators, tissue engineering,
+
                        cancer treatments, bio-computing, etc. ENABLE is a toolbox for constructing
+
                        contact-dependent signaling networks in mammals. The 3-layer design principle
+
                        underlying ENABLE empowers any future development of transmembrane logic circuits,
+
                        thus contributes a foundational advance to Synthetic Biology.

: involved, no matter the contribution is large or small

Project design »

In our brainstorm session in December 2018, Chuyue Zhang, through literal reading and practice, came up with the basic draft of the sequence-specific in vivo mutagenesis system utilizing reverse transcriptase. Zixuan Zhang and our advisor Rongrong Du offered lots of suggestions and help. In our brainstorming sessions, everyone in the team contributed to improve the project.

Prof. Liang Cai, secondary PI of our iGEM team, gave us general advice in all the aspects of our project, especially on how to make our system work better. He paid close attention to our project at every stage and offered his guidance whenever a problem was encountered.

rof. Jinzhong Lin, Prof. Hong Lv and Prof. Yongming Wang, from School of Life Sciences of Fudan University, gave us much advice on specific problems we met in the process of experiment, such as how to deal with the leakage of Cre recombinase in Cre/LoxP system, how to increase the mutation rate of reverse transcriptase and so on. They also contributed their ideas on our project's possible applications.

Prof. Alper, PI from McKetta Department of Chemical Engineering from the University of Texas at Austin, helped us improve our project design and gave us some constructive advice.

Experiments »

Chuyue Zhang took on the responsibility to manage and monitor the whole experimental group. She herself finished the construction and verification of reverse transcriptase parts. Yuchang Li was responsible for the construction and verification of the Cre/LoxP system. Fluorescent measurement was conducted by Xidan Miao. Yi Qiao was our major contributor in molecular cloning. Yichen Liu in company with Yi Qiao constructed our improved parts and measured them. Jingyu Cao also helped in molecular cloning. All the people mentioned above together finished molecular cloning. Zixuan Zhang helped us verify our experimental design.

Prof. Liang Cai let us work in his laboratory and provided us all the reagents and equipment we needed.

Prof. Daru Lu, the primary PI of our iGEM team, secured the primary funding for the team so that we got resources and consumables.

Human practice »

Yichen Liu organized most of our integrated human practice activities including communications with professors and interviews with biotech companies. Yuchang Li helped him contact professors and summarized feedback integration. Xidan Miao was in charge of our public engagement and education human practice. The events she designed and held helped arouse interest in iGEM and synthetic biology. Yichen Liu also made a contribution by designing the rules of our board game. All the art design work needed in human practice was finished by Zixuan Zhang.

Miss Dorothy Zhang, Asia Ambassador, gave us lots of instructions and suggestions on how to do HP work and shared her experience of working for former iGEM competitions. We extend our sincere appreciation to Miss Zhang for her helpful sharing!

Chen Wang, from Software School of Fudan University, helped us make a crawler program used to collect the data we need from the iGEM website.

Bozitao Zhong, our team's mentor, offered us ideas on how to conduct human practice and gave us advice on iGEM competition based on his experience.

Model »

Yuchao Huang took on the responsibility for modeling all by himself.

Yiheng Wang, from the 2017 iGEM team Fudan, gave us guidance on modeling.

Prof. Jinzhong Lin and Prof. Qiang Huang, from School of Life Science of FDU, gave us many constructive suggestions in terms of enzyme kinetics.

Software »

Yuchao Huang wrote the python program for tRNA primer designing all by himself.

Prof. Liang Cai converted the python into javascript for running online.

Hardware »

Prof. Liang Cai provided the idea and equipment for us.

Yuchao Huang optimized the original program and did the equipment test with Yuchang Li together.

Art design

Zixuan Zhang act as our sole art designer. She made the animation that introduces the principle of our system and designed team logo, slides, and posters for us to present our project. In addition, she designed our team uniforms and mascots for us, everyone.

Wiki construction

Zixuan Zhang coded the site, while the rest of the team worked on the content write-up. Xin Wang, from the 2018 iGEM Team:Fudan-China, gave us much help and guidance on wiki construction.
