Bronze Medal Requirements

  1. Registration: our team successfully registered for the 2019 iGEM Competition on 3/28/2019 as seen here.
  2. Deliverables:The wiki was updated until the freeze deadline on 10/21/19, and will be updated once more after the competition. The poster and presentation will be put on display at the Giant Jamboree.Judging form: Our judging form was completed on 10/16/19 for judges to view.

  4. Project Inspiration The story behind the inspiration for our project can be found here on our descriptions page.
  5. Characterization:Our team used part BBa_K934001 for initial PHB production to gather data on PHB quantification, characterization, and extraction methods. Our team tested and measured different culturing conditions than those initially used on BBa_K934001, Red Nile staining, and a unique method of PHB extraction using sonication and separation of cells from PHB using density differential centrifugation. The full characterization additions made to this part can be found the BBa_K934001 part site.

Silver Medal Requirements

  1. Validated Part: We have listed multiple parts we created on our parts page. One validated part would be BBa_K3192024 which is our pha plasmid that we designed to effectively produce PHB’s. We confirmed the biosynthesis of PHB from our plasmid through our sonication extraction procedure, red-nile staining, and electron-microscopy.
  2. Collaboration: Throughout the course of the project, our team initiated two collaborations on the iGEM site and completed an additional 11 collaboration requests to help out our fellow teams! More information about these projects can be found here.
  3. Our human practices efforts were focused on reforming public knowledge, as well as our own, to create a lasting impact. Our Outreach page can be found here.

Gold Medal Requirements

  1. Integrated Human Practices: Our Human Practices worked to understand the issues we were tackling and discover the impact of our device. More can be found out about this in the Integrated Practices Page.
  2. Improved a Previous Part: BBa_K3192026 is an improvement on well characterized terminator BBa_B0010, a strong terminator in the iGEM parts registry. BasI restriction sites were added to each terminator end to increase the ease of requesting this part with intentions to use Golden Gate Assembly. Noting its popular use in the iGEM competition, our team improved several parts for Golden Gate Assembly in addition to this terminator.
  3. Modeling: We built a genome-scale metabolic model to simulate our device’s theoretical maximum yield and sensitivity to medium metabolites, as well as informed the design of our implemented bioprocess. Learn more about it here.
