Escherichia coli |
Ribosome Binding Site (RBS) |
The antisense gene is flanked by an RBS sequence on the 5' end and a complementary RBS sequence on the 3' end. The RBS forms a duplex by binding to its complementary RBS sequence in the absence of antibiotic resistance gene & thus, inactivates the expression of the Cro gene. When the antibiotic resistant gene is present, the RBS binds to the complement RBS in the mRNA of the resistant bacterium & enables the expression of the Cro gene. |

Regulatory |
Escherichia coli |
Double Terminator |
The double terminator terminates the transcription of the antisense construct mRNA. |
129bp |
BBA_K3053000 |
Regulatory |
Lambda Phase |
Constitutive Promoter |
The constitutive promoter activates the transcription of all the genes in the construct. |

Coding |
Lambda Phage |
Cro Gene |
Cro protein blocks promoter of cI protein. Expression of cI protein is responsible for maintaining the phage lysogenic cycle. Therefore, expression of Cro switches the phage from its lysogenic to lytic cycle. |

Reverse Complement |
Forms a dsRNA loop in absence of antibiotic resistance gene and inhibits expression of Cro gene. |
