Week 40 (30th of September-6th of October)
3rd of October
1. Fluorescence Confocal Microscopy with Professor Alexander Schulz
Team members: Noel & Jonas
Materials and Procedure
The GPER-sfGFP and the xLHCGR-sfGFP yeast constructs were analysed under the Confocal Microscope.
Results
The constructs again showed visible fluorescence. One of the GPER-sfGFP did not show any fluorescence even though it had shown fluorescence before. This could be due to the colony being too old and not healthy anymore.
4th of October
1. Mini prep - Plasmid DNA purification
Team members: Ojas & Signe
The plasmid purification was performed on the nuclear receptor, the empty pUUS and pWUS plasmids, pWUS-TEF1-BI1 colony 2, 3,5 & 8 and pWUS-GAL1-BI1 colony 1,2 & 4.
Materials
- E.Z.N.A Plasmid DNA Mini Kit
Procedure
- We followed the protocol that came with the kit
Results
The concentration of the purified samples was measured by nanodrop.
