Introduction Parts Table Parts Characterization and Improvement

In order to conduct the necessary experiments, we designed a variety of BioBricks which were submitted to the iGEM Registry of Standard Biological Parts. Below you can find a short description of our parts. More information are available a click away; specific links lead to each BioBrick’s page.

Introduction

This year, we generated 18 new parts and integrated 7 existing parts into our project. The goal is for them to be accessible to future teams so that they can build up on them and generate completely different composite parts. For our BioBricks’ construction we made use of the IDT and Twist Biosciences synthesis offers, because of their early arrive and effectiveness. However, we used PCR to make simple changes to their sequences. Regarding the fact that reporter protein expression is a key component of our project, most of our parts are composed by the basic parts: T7 Promoter (BBa_J64997), Ribosomal Binding Site (BBa_B0034), coding sequence and T7 Terminator (BBa_K731721). Our favorite Basic part is IS6110 (BBa_K2973009) which is our biomarker. Our favorite composite part is our newly designed toehold 13 (BBa_K2973011) from the thermophilic bacterium Geobacillus kaustophilus, which is the universal sensor of our project.

Parts Characterization and Improvement

The parts we used for the characterization experiments are the TEM-optimized β-lactamase / J23119 (BBa_K2973019), the TEM-optimized β-lactamase / J23100 (BBa_K2973020), the TEM-optimized β-lactamase / J23105 (BBa_K2973021) and the TEM-optimized β-lactamase / J23106 (BBa_K2973022). The parts we used for the improvement experiments are the 32B Toehold Switch_β-lactamase_no signal peptide (BBa_K2973007), the Trigger 32B (BBa_K2973023) and the T7-RBS-β-Lactamase-No Signal peptide (BBa_K2973000). To see more about the improvement go to our Part Improvement Page.
