We have registered ourselves for iGEM, had a fantastic iGEM season and will attend the Giant Jamboree with our full team.
Competition Deliverables
We have completed the following deliverables:
- Wiki (as you can see).
- We will present our poster at the Giant Jamboree (Visit us at Zone 1, poster 60 where we will present our poster on Saturday between 1:00-2:30 pm and 6:30-8:00 pm).
- We will give our 20-minute presentation at the Giant Jamboree (Come visit us at Saturday 10 am at room 311!).
- Judging form
Attributions
We have attributed all parts of our project.
Project Inspiration and Description
We have depicted our project inspiration and description.
Characterization / Contribution
We have contributed quantitative experimental characterization data to mCherry (BBa_J06504).
Validated Part / Validated Contribution
We have documented multiple new Biobricks (BBa_K3168004 & BBa_K3168005) that function as expected and form the basis of our project by forming the paired dCas9-Split-NanoLuc system.
Collaboration
We have participated in several collaborations, ranging from fun collaborations to content-related collaborations and organized a BeNeLux Mini Jamboree a week prior to the Giant Jamboree.
Human Practices
We have investigated and clearly documented the impact of our project on various stakeholders.
Integrated Human Practices
We have integrated feedback from stakeholders and experts into our project and design and documented this on the Integrated Human Practices page.
Improve a Previous Part / Project
We have improved NanoLuc (BBa_K1159001) by creating a cysteine-free-NanoLuc version (BBa_K3168006) that enables usage in fusion proteins that require to be labeled with maleimide coupling, such as our own dCas9-BRET (BBa_K3168007).
Model Your Project
We have developed a model that can be used as a tool to find the right gRNA that is needed in our detection system. At the same time, we have modelled the interaction between bacteriophages and bacteria to optimize the conditions for our detection system.
Demonstration of Your Work
We have demonstrated that our detection system works under realistic conditions.
