Our part Laccase (lcc1) BBa_K2962000 was characterized by iGEM10_Tianjin (BBa_K500002). This Trametes versicolor laccase gene (Laccase (lcc1) BBa_K2962000) is an optimized sequence for expression in the E. coli organism. To avoid codon degeneracy, we innovated a new sequence. The degeneracy of the genetic code is valuable to living things because it allows for more than one codon to code for one amino acid. These characteristics(codon degeneracy) of the genetic code make it more tolerant to unwanted mutations. For example, theoretically, four-fold degenerate codons can be expected to be resistant to a mutation in the third position (but in fact codon usage bias limits this in most organisms).

We designed all of our other parts according to this laccase.

Our Pelb signaling sequence (BBa_K2962004) was characterized by BBa_K1127003 by team iGEM13_York_UK, and it was used for the periplasmic localization of the laccase enzyme in E. coli. This is expected to increase the yield of purificated protein. We innovated a new gene sequence which is compatible with our laccase. It means we used an appropriate PelB signal sequence for our laccase enzyme.
