Part Collection - STE2, more than one-trick pony
We provide a coherent platform merging directed evolution of GPCRs with the various mating reporters to enable the conversion of yeast into customised live-cell peptide sensors. We built a variety of innovative reporter constructs based on GPCR-ligand interactions around the mating receptor of mating type a (MATa) yeasts STE2. Here we present several key components of our work focussing on STE2:
The ace of the show. Tainted by a PstI cutsite and biobrick uncompatible it was not the first tier choice looking for useful parts in the registry. Sometimes a short makeover can have a huge impact: we can now present a yeast and biobrick optimized STE2 expression plasmid: BBa_K3173005.
STE2 can reliably propagate a binding system inside the cell starting a highly researched signalling cascade.
Make it chimeric
Exchanging the binding pocket to program a new input: We exchanged the middle part of STE2 for a fractionof the human GPCR FPR2 (find out more about our collab with Freiburg:Collaborations) This chimeric design is based on the DIY receptor design from iGEM TU-Delft 2012.
