- Made the PA medium for the plastic experiments
- Cultured the 20Z and put in test tubes with the plastic to see how the plastics would react
- Cut the plastics and insert them into the tubes with the inoculated bacteria

- Made the PA medium for the fourth plastic
- Cultured the 20Z and put in test tubes with the plastic to see how the plastic would react
- Took ODs of the plastics to measure growth of the bacteria
June
Week 3

Monday
- Finished the experiment involving the plastics to see which interacted well with the 20Z
- Took last ODs of plastics
- Took plastics out of test tubes and placed in dry area over night
- Printed surveys and brochures to pass out
Tuesday
- Visited Granger Landfill Facilities
- Weighed the plastics, recorded which ones gained mass, meaning a biofilm was formed
- Resin seemed to do the best and a biofilm was formed, future testing will be done with different types of resin with different pigmentation

- Ordered more 3D prints for testing
- Worked on wiki interface

- Inoculated each tube
- Added the 5 types of Resin plastic (black, white, clear, high temperature, dental)
- Recorded the OD at hour 0 and shook half (two test tubes of each type of plastic) overnight at 30 ºC, other half (two test tubes of each type of plastic) in a stationary area

- Took ODs every two hours
- Skyped with UT Austin’s iGEM team about their project and collaborating

- Continued taking optical density every two hours
- Worked on Wiki Interface

- Continued taking ODs of the test tubes
- Met with a CSE professor for help with coding the Wiki

Tuesday
- Took final optical densities
- Took plastics out of test tubes and put them in 30 degree incubator overnight to dry
- Met with someone in the engineering department to talk about sponsorships and possible mentors
Wednesday
- Took plates out of the incubator
- Weighed the plastics
- High temperature and dental resin gained mass, meaning a biofilm was formed
- Future work will be done with these two plastics
Thursday
- Worked on Wiki Interface
- Started writing the children's book
Friday
- Contacted landfills about visiting and asking questions

- Ordered new plastics for testing
Wednesday
- Made MPM media solution
- Weighed plastics, using black resin, white resin, dental resin, high temperature resin
- Added plastics to the test tubes
- Inoculated the wildtype AM1 culture
- Add 2 mL of sterile water to a flask
- Let freezer stock of the wildtype AM1 culture melt
- When melted, pipet about 500 uL of the stock into the flask
- Put 150 uL of the bacterial solution into each test tube
- Took first optical density at time 0 hrs and put tubes in the shaker
- Continued taking optical densities every two hours
- Contacted several professors about meeting up to discuss the bioreactor and funding

- Added 250uL methanol to the tubes, 750 uL at end of day for weekend
- Created a new tube to take second OD’s then added more methanol
July
Week 7
06/30 - 07/06
Monday
- Added 250 microliters of methanol to all the tubes
- Took OD’s in new diluted tubes
Tuesday
- Took OD’s, did not refeed
- Made more MP media
- Met with Professor Wei Liao about the design and construction of our bioreactor
Wednesday
- Began to investigate mass transfer for the methane capture system
- Each member read through the “Systematic metabolic engineering of Methylomicrobium alcaliphilum 20Z for 2,3-butanediol production from methane” paper
- Took last optical densities
- Started harvesting cells from MPM culture
Week 8
07/07 - 07/13
Monday
- Took final mass of plastics to compare to initial mass
- Took mass of harvested cells
- Made MPM media
- Combined P medias
- Inoculated culture into the MPM and P medias
- Ordered 20 honeycomb design plastics to use for testing (10 in high temp and 10 in dental)
- Discussed with Methane regulation people
Tuesday
- Inoculated the bacteria
- Worked on researching bioreactor design
Wednesday
- Got 3D prints, cut out inserts
- Helped with the UT Austin Burden Assay collaboration
- Took optical density
- Took 50 uL of the PA1 bacteria and put into 3 mL of MP Media
- Put back on shaker with 20Z bacteria
Thursday
- Took optical density
- Made P Media agar
- Same protocol as 200 mL P Media solution but added 2.4 g of agar before autoclaving
- Finished MPM agar
- Warmed it up for 5-7 minutes at 50 power
- Added 1 mL of methanol
- Dipped each plastic in methanol and then put on plate
- Poured P Media agar and MPM agar into plates
- 10 plates each, let set for an hour
- Pipette 10 uL of bacteria into each hexagonal cell in the plastic (7 on each)
- 10 plates of 20Z and 10 plates of PA1
- Put into 30 degree incubator to grow

- Re-fed PA1 with 10 uL of methanol
- Researched bioreactor
- Worked on Wiki
Tuesday
- Re-fed PA1 with 10 uL of methanol
- Researched bioreactor
- Re-fed PA1 again with 50 uL of methanol on plastic
Wednesday
- Handed out surveys
- Made MPM solution using 15 mL of MP Media and 75 uL of methanol
- Pipetted 1 mL on to each PA1 plate
- Ordered more 3D prints, 5 of dental and 5 of high temperature
Thursday
- Met with Wei Liao to talk about bioreactor
- Re-fed PA1 with 15 uL of MPM solution for each cell, and some on plastic
Friday
- Went to Waste Management in lennon to talk to Rodney about bioreactor and system
- Further research on our discussion
- Re-fed PA1 with 15 uL of MPM solution for each cell, and some on plastic
- Weighed new plastics for initial mass

- Poured the plates and set plastics in them

- Re-fed PA1 plates with 15 uL of MPM solution for each cell, and some on plastic

- Presented our research findings at MSU’s 2019 Mid-Michigan Symposium for Undergraduate Research Experiences (Mid-SURE)
