Safety
AHUT_China team attaches great importance to safety issues. Before officially entering the laboratory to conduct experiments, each of us carefully read the documents related to laboratory safety and conducted laboratory biosafety training. The members were not allowed to conduct experiments until they passed the safety assessment.
General Laboratary Satety
When we work in the lab, we wear appropriate overalls or protective clothing and appropriate glove.
Personnel unrelated to the work of the laboratory shall not enter the laboratory without authorization. If visitors from other units need to enter the laboratory, they can only enter the laboratory after obtaining the approval of the person in charge of the laboratory.
We wash our hands carefully after the experiment, and check carefully when we leave the lab.
We don't eat, drink, smoke, make up or handle contact lenses in the lab work area.
All drugs and reagents have a designated location, the use and purchase of drugs and reagents should be placed in the designated location, all kinds of drugs and reagents should have correct and clear label, including name, concentration, specifications, etc., according to the correct way to use.
Our laboratory carries out strict hygiene responsibility system, and the indoor sanitation will be cleaned regularly to keep the laboratory table and fume hood clean and tidy.
Project Specific Laboratory Safety
First, our experiments do not involve any experiments of human, animal or plant and are in full compliance with iGEM's safety and security policies.
Second, the laboratory we are in is a low-risk laboratory (level 1), and the specific experiments of the project are also completed in safe and clean laboratory bench and ultra-clean workbench.
Third, as our experimental chassis, E. coli is one of the most widely used safety strains in synthetic biology and does not cause any harm to human or the environment.
Fourth, all kinds of reagents used in the experiment are harmless, and the waste liquid generated from the experiment will be treated normatively to ensure that it will not harm the environment after being discarded.
Our laboratory waste treatment requirements
Laboratory waste disposal
Solid waste
Solid culture medium containing bacteria and disposable waste such as gun head, plastic pipe, gloves, hats, masks, etc. are put into special garbage bags and burned. Reusable glass devices, such as test tubes, glass bottles, or plastic products, can be soaked with lionser disinfection tablet solution (3 tablets /1000ml water, soaked for more than 3 hours), then cleaned and used after high-pressure steam sterilization.
Liquid waste A liquid culture containing bacteria
After adding lionser disinfection tablet to deal with (5 piece/1000ml H2O, soak for more than 3 hours. According to the concentration of bacteria, the dosage of disinfection tablets can be increased or the soaking time can be prolonged) or boil the liquid containing bacteria directly for 30min, the liquid culture containing bacteria can be poured down the drain. Do not directly dispose of bacteria-containing waste into the sewer.
Pipette Tips
The used pipette suction head should be soaked in a container containing 10% sodium hypochlorite solution for not less than 24 hours.
PCR product
All liquids and wastes containing PCR products should be soaked in a container containing 1mol/L HCI, and the soaking time should not be less than 6h. While using the way of PCR-ELISA , the waste liquid produced by Microplate Washer should be collected into 1 mol/L HCl solution.
Positive samples and plasmids
The positive plasmid should be soaked in 1mol/L HCl solution, and the soaking time should not be less than 6h.
