1. REPETITION OF COLONY PCR

The colony PCR was done again on as problems appeared in the colony PCR done in the last couple of days. The colony PCR was done on the following:

*The positive control was from the kill switch group. This was used as we knew the colony was positive and should therefore show up on the gel if everything worked as it should.

Procedure
- The colony PCR was done according to the normal procedure, but we tried to use MangoMix instead of making normally mastermix.
- 3 colonies was taken pr. Plates* (only one colony from the positive control plate)
- *one sample with colonies from plate 7 went wrong and we did not use if further on.
- Loading order: 4,4,4,7,7,3,3,3, negative control, positive control (colony 8 on plate X12A-TEF1+BAX from kill switch work).

1. USER LIGATION
Team members: Swenja and Noël
We redid the USER ligation because the colony PCR on the 19/8 didn’t show desired bands. Procedure as on the 20/8.
