Overview
We designed a new part (pOmpC-TRZ1-iLOV) to evaluate the impact in cell viability of the overstimulation of the endogenous EnvZ/OmpR system, through the establishment of a positive feedback loop between the TRZ1 receptor and the pOmpC.

•
pOmpC/GFP
- This part shows the activity level of pOmpC.
- Depending on the activity of pOmpC, the expression level of GFP changes in a directly proportional way, giving up a measurable fluorescent signal.
•
pBAD/TRZ1 Part number: BBa_K3149000
- Codes for a membrane protein that recognizes glucose bound to GBP (glucose binding protein).
- pBAD is an inducible promoter that allows TRZ1 expression according to the available arabinose concentration.
- Detects the complex formed by glucose and ribose with their respective binding proteins and signals the EnvZ domain to signal the osmoporin downstream signalling pathway.
•
pOmpC-TRZ1-iLOV Part Number:BBa_K3149001
- Codes for the osmolarity-sensitive pOmpC, along with the aforementioned TRZ1 protein along with a fluorescent reporter (iLOV).
- This allows both the expression of TRZ1 and the expression of iLOV and its fluorescent signal according to osmolarity changes in the extracellular media.
