Team:Tartu TUIT/Parts
This year our team has contributed 17 diverse parts to the registry: 2 Regulatory, 4 Protein_Domain and 11 Coding.
One of the most crucial parts of our project was the implementation of yeast secretion signal: from alpha-factor peptide pheromone and Ost1 protein. Our team used them to enhance the secretion of the produced enzymes. Their sequences were added to the registry as Protein_Domain parts. We have also added their modified versions fused with the HA tag, that we were using for the Western Blot.
Coding parts are S. cerevisiae gene (SCW4) and codon-optimised for S. cerevisiae A. luteus gene (GLC1) coding for glucanases.
For GLC1 we have also added the altered versions, fused with the secretion signals.
We have also added the improved versions of BBa_K2711000 part, added by UiOslo_Norway 2018 team. This is an A. luteus gene coding for glucanase. Our team has managed to enhance its activity by the addition of the secretion signals.
As Regulatory parts, our team has added two S. cerevisiae promoters: pRNR1 and pGAL1.
Overall, this year we have managed to use a wide range of different parts, that we have also successfully characterized.
