We were trained with synthetic biology knowledge and experimental technology by our instructor Liu Xinyu.
We reviewed and discussed some previous iGEM projects.
We brainstromed in groups, discussed and proposed several doable projects.
We conducted paper researches, and finally determined the theme of our project.
We designed our project with the help of our instructor Meng Fankang.
June~October
We conducted the experiments.
Week1
We got MerR and pMerR from iGEM kits, and plasmid backbones from Institute of Microbiology, Chinese Academy of Sciences.
Week2
We contructed the basic Hg sensor J23101-MerR-pMerR-GFP
We modeled the effect of the constitutive promoter strength to the basic Hg sensor.
Week3
We contructed a series of basic Hg sensors with different J23 promoters.
Week4~6
We got INP, SpyCatcher, SpyTag and CsgA from iGEM kits.
We constructed CsgA-INP-SpyTag.
Week7~8
We tested the efficiency of the basic Hg sensor.
We deleted the csgA gene in E.coli C321 strain.
Week 9
We got the amplifer RinA_P80α from QHFZ_2019, and TEV-C1434 from UCAS_China.
Week 10~11
We designed and constructed the improved version of amplifer, which combined RinA_P80α and TEV-C1434.
Week 12
We tested the efficiency of the three amplifers on our Hg sensor.
Week 13~14
We constructed SpyCatcher-MBP and expressed the protein.
Week 15~16
We synthesized and constructed the parts used in our biocontainment module.
Week 17~18
We tested the effect of our biocontainment module.
Week 19~20
We tested the effect of our CsgA-INP-SpyTag with SpyCatcher-MBP system to the absorption of Hg2+.
