Parts
This year, iGEM Bulgaria has submitted the following parts:
- BBa_K3208000 a composite part that is an expression cassette for heterologous production of AMPs in E. coli. It contains an N-terminal DAMP4 tag for efficient expression and simple purification of the AMPs. The cloning of new peptide sequences can be done using the two Eco31I sites that allow scarless insertion.
- BBa_K3208001 to BBa_K3208009 – expression modules for putative antimicrobial peptides isolated from the genome of the great white shark (Carcharodon carcharias).
- BBa_ K3208010 a red expression cassette that can be inserted between two Eco31I sites. This modification can significantly decrease the number of colonies that have to be screened to find a correct transformant when using vectors that rely on Eco31I digestion and ligation.
- BBa_ K3208011 a modified version of part BBa_K3208000 with part BBa_ K3208010.
- BBa_ K3208012 improved version of part BBa_K2515002 from 2017. The old gRNA cloning vector is modified in a way to contain a red expression cassette between the two Eco31I sites. This modification significantly decreases the number of colonies that have to be screened to find a correct transformant.
