  1. In previous work of screening for autophagy agonists, several autophagy-related protein such as LC3, p62 and Beclin 1 were detected to evaluate the development of autophagy by western blot assay. However, as a dynamic degradation process in cell, it is hardly to assess autophagy process accurately through detecting certain autophagy protein. In this work, we evaluated the autophagy process through autophagic flow assay, a more intuitive assay to monitor the dynamic process of autophagy including formation autophagosome and fusion of autophagosomes and lysosomes.   2. Given that monitoring autophagic flux is important for the analysis of autophagy, an mTagRFP-mWasabi-LC3 reporter was used in our screening platform. Compared with the superannuated Tandem fluorescent-tagged LC3 (mRFP-EGFP-LC3), mTagRFP-mWasabi-LC3 is more sensitive and accurate in detecting the accumulation of autophagosomes and autolysosomes which due to mWasabi is more acid sensitive than EGFP and has no fluorescence in acidic lysosomes. In view of the advantages of “New Uses of Old Drugs” in accelerating the new drugs development speed and saving research and development costs, we screened for autophagy agonist from FDA-approved drugs and attempted to find compounds with anti-AD activity from existing clinical drugs.
