prmA promoter BBa_K2911000 This promoter comes from the prmA gene sequence. The prmA gene is a stress regulatory gene that was found to be regulated in the presence of PFAAs, harmful chemicals that have been found in water supplies around the world. Once turned on, the prmA gene is found to induce floculation as a stress response. This part is the isolated promoter taken from this gene. Previous studies show a 3-fold upregulation of the stress-associated gene, prmA, in Rhodococcus jostii RHA1 when grown in 100uM PFOA.
Rhodococcus Plasmid pSRKBB BBa_K2911001 Rhodoccocus Plasmid pSRKBB. This plasmid consists can be used in both E. coli and Rhodoccocus. This plasmid has multiple cut sites including; SpeI, EcoRI, PstI. Origin of replication pSKBB-empty [4566-5141]. Antibiotic resistant gene NeoR/KanR.
Insert prmA/mRFP BBa_K2911002 Insert includes promoter region identified in Rhodococcus RHA1.This promoter region is responsible for flocculation in Rhodococcus as a stress response. The insert also includes mFRP part BBa_E1010, a ref fluorescent protein with increased transcription due to the promoter sequence. The insert includes the promoter region, prm A, identified in Rhodococcus RHA1
pSRKBB/prmA/mRFP BBa_K2911003 The pSRKBB-prmAmRFP composite plasmid is can be transofrmed into E. coli and Rhodococcus jostii RHA1. In RHA1, this part serves as a reporter that glows red in response to PFOA and PFOS exposure.
