MEASUREMENT

For creating our diagnostic qualitative signal model, we had to utilize available information regarding the reporter protein, amajLime, and determine its signal intensity from measurements taken in our lab. The essential measurements for the model are for the expression of amajLime in known quantity of the reporter protein in the growth curve and the crude detection solution. Another essential aspect of the model is the paper assay that was performed to determine ideal paper and solvent conditions. Solvents based on water, organic and detergent based solutions were used to measure the movement of the amajLime protein band. The paper was selected for the quickest speed of chromatography and separation of reporter protein added to the crude sample. After the appropriate paper was selected, the solvent was selected to provide the fastest chromatography result and band intensity. The measurements performed for chromatography were used to create the training kit for the detection system.
