A BRIEF INTRODUCTION

For the first time, Tel Aviv University will be participating in the iGEM competition. Most of our work was done independently by the team members with guidance from our PIs and Instructor. This includes planning the biological constructs, raising external funds and supplies, designing the website and executing the lab work.

The iGEM course and team first began its work in the university in late 2018. The course was divided into two parts: during the first semester we received general theoretical classes in our PIs fields of study as well as an introduction to synthetic biology by Dr. Johann Elbaz.

During the final weeks of our first semester, we reviewed past iGEM projects and brainstormed our own project and field of interest. In January, we finalized our topic for the project.

Starting in March 2019, we have self-studied the theory behind cloning and plasmid planning. Furthermore, we have gone through a 2-day seminar about cloning and making competent bacteria.

The rest of the work was mainly completed by the team, as stated above. BSL2 procedures were consistently done under the inspection of the Salomon lab.

SCIENTIFIC ATTRIBUTIONS

Wet lab Guidance:

Dr. Dor Salomon (Secondary PI) Dr. Salomon helped us find our way through the microbiology and theoretical articles. He constantly assisted in planning almost every part of the project and contributed in analyzing the results as well.

In addition, Dr. Salomon hosted the team at times when a BSL2 level experiment was performed.

Dr. Johann Elbaz (Secondary PI) Dr. Elbaz introduced us to the field of synthetic biology, and allowed for innovative thinking. He also inspired us to look for alternatives when we struggled to assemble the pyocin cluster via Gibson assembly, later instructing and assisting us with the Golden Gate method.

Mr. Dan Alon (Instructor) Dan Alon was available for all of our questions regarding protocols and cloning troubleshooting. One specific and special donation Dan made to the project is working with us while adapting our Cluster sequences to the Golden Gate assembly method.

Modeling and Bioinformatics:

Prof. Tamir Tuller (Primary PI) Prof. Tuller instructed both our Engineering and Bioinformatics team members, giving them advice on any matter related to the computational parts of the project. In addition, Professor Tuller assisted us with issues of funding and financial logistics.

DESIGN

Site Design - Wiki design and temporary site design was don solely by team members Lilach Elbaum and Lior Soffer.

Logistics

Dr. Anat Parket Dr. Parket allowed us to have a bench in the educational laboratory, but most importantly, forgave us even though we forgot to close the -20 degrees fridge, causing all its content to defreeze.
